A postdoctoral position is available in the Center for Immunology and Inflammatory Diseases at Massachusetts General Hospital – Harvard Medical School to study CD4+ T cells in Rheumatoid Arthritis (RA) and Lyme arthritis (LA), a tick-borne infection that may lead to autoimmune, chronic inflammatory arthritis. The position is part of recently funded collaborative project between the laboratories of Dr. Allen Steere, who studies human immunology of RA and LA, and Dr. James Moon, who studies T cell tolerance to commensal bacteria and self-antigens. Studies will address the hypothesized role of T cell immunity to commensal bacteria in driving the pathogenesis of RA, and T cell immunity to the Lyme Disease pathogen Borrelia and related autoantigens in driving the pathogenesis of LA. Experimental strategies will focus on the use of custom peptide:MHC tetramer technology to directly identify and characterize CD4+ T cells with specificity to commensal bacteria species associated with RA, pathogenic Borrelia species involved in Lyme disease, and autoantigens targeted in RA or LA. A variety of methodologies will be employed in conjunction with tetramers to study antigen-specific T cells from the blood and synovial fluid of RA and LA patients, including advanced flow cytometry, the generation of clonal T cell lines, and in particular, single cell RNAseq transcriptomics and TCR repertoire analysis in conjunction with the center’s Single Cell Genomics Program.

Candidates must be highly motivated, well organized, technically proficient, critically minded, and able to work independently but collaboratively with others. Candidates must possess a recent PhD, MD, or MD/PhD with a strong background in cellular or molecular immunology. Extra consideration will be given to those with relevant experience with flow cytometry, in vitro T cell assays, and systems biology.
